Pemazyre is a fibroblast growth factor receptor (FGFR) inhibitor indicated for the treatment of adults with previously treated, unresectable locally advanced or metastatic cholangiocarcinoma with FGFR2 fusion or rearrangement.
Recommended Dosage:
- 13.5 mg orally once daily for 14 days, followed by 7 days off (21-day cycle)
- Continue until disease progression or unacceptable toxicity
- Tablets should be taken whole, with or without food, around the same time daily
- Do not crush, chew, split, or dissolve
- Missed dose/vomiting: Do not retake; continue with next scheduled dose
Eye Toxicity (RPED): May cause vision changes such as floaters or blurring. Perform baseline and periodic ophthalmic exams with OCT
Hyperphosphatemia: Monitor serum phosphate. Begin a low phosphate diet for levels > 5.5 mg/dL and initiate medical intervention for levels > 7 mg/dL
Fetal Harm Risk: May cause fetal harm. Advise use of contraception during treatment and for 1 week after final dose (both females and males with partners of reproductive potential)
