Zydelig (Idelalisib) is a selective oral phosphoinositide 3-kinase delta (PI3Kδ) inhibitor used in the treatment of hematological malignancies.
Indications:
Zydelig is indicated for use in the treatment of the following:
- Chronic Lymphocytic Leukemia (CLL): Used in combination with rituximab in relapsed CLL for patients who are not suitable for rituximab monotherapy due to comorbidities.
- Follicular Lymphoma (FL): For relapsed cases where patients have received at least two prior systemic therapies.
- Small Lymphocytic Lymphoma (SLL): Indicated in patients who have relapsed after two or more systemic therapies.
Recommended Dosage:
Typically prescribed as 150 mg taken orally twice daily. The dosage may be adjusted based on patient tolerance and adverse reactions. Treatment continues until disease progression or unacceptable toxicity occurs.
Serious and potentially fatal hepatotoxicity, colitis, pneumonitis, and intestinal perforation have been reported. Monitor liver function and respiratory symptoms closely.
Infections (including opportunistic infections such as PJP) may occur. Prophylaxis and regular clinical assessments are recommended.
Severe diarrhea and colitis can arise during treatment. Discontinue Zydelig if severe gastrointestinal symptoms are present.
Avoid use in pregnancy; Zydelig may harm the unborn baby. Women of childbearing potential must use effective contraception during and for 1 month after treatment.
Do not breastfeed while taking Zydelig and for at least 1 month after the last dose.
