Daybue is a prescription oral therapy used for the treatment of Rett syndrome in adults and pediatric patients aged 2 years and older.

Recommended Dosage:

  • Administer twice daily, in the morning and evening

  • Can be taken with or without food

  • Administered orally or through a G-tube (use G-port for GJ-tubes)


Weight-Based Dosing:

  • 9–<12 kg: 5,000 mg (25 mL) twice daily

  • 12–<20 kg: 6,000 mg (30 mL) twice daily

  • 20–<35 kg: 8,000 mg (40 mL) twice daily

  • 35–<50 kg: 10,000 mg (50 mL) twice daily

  • ≥50 kg: 12,000 mg (60 mL) twice daily
  • Diarrhea: Most patients may experience diarrhea. Cease laxatives before starting Daybue. Treat with antidiarrheals, hydrate, and monitor symptoms. Adjust or discontinue dosage if severe.

  • Weight Loss: Monitor weight regularly. Interrupt or reduce dose if significant weight loss is observed.

  • Common Adverse Reactions: Diarrhea and vomiting are the most frequently reported side effects.

Drug Interactions:

  • Monitor for interactions with CYP3A4-sensitive substrates

  • Avoid concurrent use with OATP1B1 and OATP1B3 substrates that may cause toxicity with altered plasma concentration
