Vitrakvi is an oral kinase inhibitor indicated for the treatment of adult and pediatric patients with solid tumors that:
- Exhibit NTRK gene fusion (without known acquired resistance mutations)
- Are metastatic or where surgical resection is likely to cause severe morbidity
- Have no satisfactory alternative treatment options or have progressed following prior therapy
Recommended Dosage
- Adults and pediatric patients with BSA ≥1.0 m²: 100 mg twice daily
- Pediatric patients with BSA <1.0 m²: 100 mg/m² twice daily
- Take orally, with or without food
- Swallow capsules whole. Do not chew, crush, or break
- Do not retake a missed dose within 6 hours of the next scheduled dose
- If vomiting occurs, resume with the next scheduled dose
- Central Nervous System Effects: May include mood changes, dizziness, sleep disorders, or cognitive impairment. Avoid driving or operating machinery if symptoms occur.
- Fracture Risk: Monitor for pain or mobility changes. No conclusive data on bone healing.
- Liver Enzyme Elevation: Monitor AST/ALT levels every 2 weeks during the first month, then monthly.
- Pregnancy Risk: Larotrectinib may cause fetal harm. Use effective contraception during treatment and for 7 days after the last dose.
