Asparlas is an asparagine-specific enzyme used as part of a multi-agent chemotherapy regimen for the treatment of acute lymphoblastic leukemia (ALL) in pediatric and young adult patients aged 1 month to 21 years.
Recommended Dosage:
- 2,500 units/m² administered intravenously no more than once every 21 days
- Premedication with acetaminophen, diphenhydramine (H1-blocker), and famotidine (H2-blocker) is recommended 30–60 minutes prior to reduce the risk of infusion and hypersensitivity reactions
Hypersensitivity Reactions: Grade 3/4 reactions, including anaphylaxis, may occur. Monitor for swelling, rash, bronchospasm, and hypotension.
Pancreatitis: May be life-threatening. Monitor amylase/lipase levels. Discontinue immediately if suspected.
Thrombosis: Serious thrombotic events have been reported. Discontinue in such cases.
Hemorrhage: Monitor for increased PT, PTT, and hypofibrinogenemia; assess coagulation parameters if bleeding symptoms occur.
Liver Toxicity: Monitor bilirubin and transaminases weekly during treatment and up to 6 weeks after the last dose.
Pregnancy Risk: May cause fetal harm. Conduct pregnancy testing before initiation. Avoid breastfeeding during treatment and for 3 months after the last dose.
