Ayvakit is a kinase inhibitor approved for the treatment of:
- Gastrointestinal Stromal Tumor (GIST): For adults with unresectable or metastatic GIST with PDGFRA exon 18 mutations, including D842V
- Advanced Systemic Mastocytosis (AdvSM): Includes mast cell leukemia, aggressive SM, and SM-AHN (associated hematological neoplasm)
Recommended Dosage:
- GIST: 300 mg orally once daily
- AdvSM: 200 mg orally once daily
- Take on an empty stomach (≥1 hour before or ≥2 hours after meals)
- Do not retake if vomiting occurs; resume at next scheduled dose
- Avoid taking a missed dose if <8 hours remain until the next one
Intracranial Hemorrhage: Risk is higher in patients with thrombocytopenia or recent cerebrovascular events. Monitor carefully.
Platelet Count: Not recommended for AdvSM patients with platelet count <50 × 10⁹/L.
Cognitive Effects: May include memory loss or confusion. Temporarily withhold or reduce dosage based on severity.
Fetal Risk: Can cause harm to an unborn baby. Use effective contraception during treatment and for 6 weeks post-treatment.
Breastfeeding: Avoid during treatment and for 2 weeks after the final dose.
