Empaveli is a complement inhibitor used for the treatment of adults diagnosed with paroxysmal nocturnal hemoglobinuria (PNH), a rare blood disorder that can lead to hemolysis (destruction of red blood cells).

Recommended Dosage:

  • Patients must receive vaccinations against encapsulated bacteria (Neisseria meningitidis, Streptococcus pneumoniae, and Haemophilus influenzae type B) at least 2 weeks before starting Empaveli.

  • If immediate therapy is required, vaccines may be administered promptly, along with a 2-week course of antibiotics.

  • The standard dose is 1,080 mg via subcutaneous infusion twice weekly, delivered through an infusion pump with a minimum 20 mL reservoir.


If a dose is missed, administer it as soon as possible, and then resume the regular schedule.

  • Severe Infections: Use of Pegcetacoplan may increase the risk of life-threatening infections caused by encapsulated bacteria. Monitor closely for early signs of infection.

  • Vaccination Requirement: Vaccination is mandatory before starting treatment. If therapy begins before vaccination, antibiotic prophylaxis is required.

  • Hypersensitivity Reactions: Systemic allergic reactions, including anaphylaxis, may occur. Stop treatment and initiate appropriate medical care if symptoms develop.

  • Post-Treatment Monitoring: After stopping Empaveli, monitor patients for at least 8 weeks for signs of hemolysis or recurrence of PNH symptoms.

  • Pregnancy and Breastfeeding: Women of childbearing potential should use contraception during and for 40 days after treatment. Avoid breastfeeding during and for 40 days post-therapy.
