Exkivity is an oral tyrosine kinase inhibitor (TKI) indicated for the treatment of adult patients with:
- Locally advanced or metastatic non-small cell lung cancer (NSCLC) with EGFR exon 20 insertion mutations,
- Whose disease has progressed on or after platinum-based chemotherapy
Recommended Dosage:
- 160 mg orally once daily, with or without food, until disease progression or unacceptable toxicity
- Swallow capsules whole without chewing, opening, or dissolving
- If a dose is missed by more than 6 hours, skip and take the next dose as scheduled.
- If vomiting occurs, do not retake the dose. Continue with the next scheduled dose.
QTc Prolongation & Torsades de Pointes: May be fatal. Evaluate and correct electrolyte imbalances before initiating therapy. Monitor ECG and electrolytes regularly.
Interstitial Lung Disease (ILD)/Pneumonitis: Potentially fatal. Monitor for pulmonary symptoms. Discontinue permanently if ILD/pneumonitis is confirmed.
Cardiac Toxicity: May lead to heart failure. Assess left ventricular function periodically. Adjust dosage or discontinue based on severity.
Severe Diarrhea: Initiate antidiarrheal treatment at first sign. Monitor hydration and electrolytes. Adjust dose if diarrhea is severe.
Pancreatic Toxicity: Monitor amylase and lipase levels regularly, especially in patients with a history of pancreatitis.
Pregnancy & Breastfeeding Risk: May cause fetal harm. Verify pregnancy status before starting. Avoid breastfeeding during treatment and for 7 days after final dose.
