Idhifa is an isocitrate dehydrogenase-2 (IDH2) inhibitor indicated for the treatment of adult patients with relapsed or refractory acute myeloid leukemia (AML) who have a confirmed IDH2 mutation.

Recommended Dosage:

  • Treatment should begin only after confirming the presence of IDH2 mutations in the blood or bone marrow.

  • The recommended dose is 100 mg orally once daily, with or without food.

  • Tablets should be swallowed whole and should not be split or crushed.

  • Treatment may continue until disease progression or intolerable side effects. Patients showing no progression or toxicity should continue therapy for a minimum of six months to assess response.

  • If a dose is missed or vomited, take it as soon as possible on the same day and resume the regular dosing schedule the following day.
  • Monitoring: Regularly monitor blood counts and chemistries, especially during the first 3 months of therapy, to manage potential leukocytosis and tumor lysis syndrome.

  • Differentiation Syndrome: Idhifa may cause life-threatening differentiation syndrome. Initiate corticosteroids and close monitoring if symptoms develop. Hospitalization may be necessary.

  • Reproductive Risk: Idhifa may cause fetal harm. Both male and female patients of reproductive potential should use effective contraception during treatment and for at least 30 days post-therapy.

  • Breastfeeding: Women should avoid breastfeeding during treatment and for 30 days after the last dose due to potential risk to the infant.
