LORBRENATM (Lorlatinib) is a third-generation ALK inhibitor used for the treatment of adult patients with ALK-positive metastatic non-small cell lung cancer (NSCLC).
Recommended Dosage:
- 100 mg taken orally once daily, with or without food
- Tablets should be swallowed whole—do not crush, chew, or split
- If a dose is missed, it can be taken unless the next scheduled dose is due within 4 hours
- Do not take an extra dose if vomiting occurs after taking a dose; continue with the next scheduled dose
Treatment is continued until disease progression or unacceptable side effects occur.
CNS Effects: May include seizures, psychosis, cognitive changes, mood alterations, and sleep disturbances.
Hyperlipidemia: Monitor serum cholesterol and triglycerides prior to starting and during treatment.
Heart Conduction Abnormalities: QT prolongation, PR interval changes, and AV block have been reported. ECG monitoring is recommended.
Interstitial Lung Disease (ILD)/Pneumonitis: Evaluate patients with new respiratory symptoms.
Hypertension: Monitor blood pressure periodically during treatment.
Reproductive Toxicity: Lorlatinib may cause fetal harm. Women of childbearing potential must use effective non-hormonal contraception during therapy.
