Piqray is a PI3K-alpha-specific kinase inhibitor used in combination with Fulvestrant to treat:
- Postmenopausal women and men
- With HR-positive, HER2-negative, PIK3CA-mutated, advanced or metastatic breast cancer
- Whose disease has progressed after endocrine therapy
Recommended Dosage:
- 300 mg orally once daily (2 × 150 mg tablets) with food
- Fulvestrant to be administered per standard protocol (500 mg on Day 1, Day 15, Day 29, then monthly)
- Continue treatment until disease progression or unacceptable toxicity
- Do not crush or chew tablets. If a dose is missed by more than 9 hours or vomiting occurs, skip and resume the next day
Severe Allergic Reactions: Including anaphylaxis. Discontinue immediately if symptoms appear
Cutaneous Reactions: Rare but serious conditions such as SJS, TEN, and DRESS have been reported
Hyperglycemia Risk: Monitor FPG and HbA1c before starting and regularly thereafter. Manage proactively
Pneumonitis/ILD: Discontinue permanently if confirmed
Gastrointestinal Toxicity: Diarrhea and colitis-like symptoms may lead to dehydration or kidney issues
Pregnancy Risk: Can cause fetal harm. Use contraception during treatment and for 7 days post-treatment
