 Rozlytrek is a kinase inhibitor approved for the treatment of:

  • Adults with metastatic non-small cell lung cancer (NSCLC) whose tumors are ROS1-positive

  • Adults and pediatric patients (≥12 years) with NTRK gene fusion-positive solid tumors without a known acquired resistance mutation, who have progressed on prior therapy or have no satisfactory alternative options

Recommended Dosage:

  • ROS1-positive NSCLC (Adults): 600 mg orally once daily with or without food

  • NTRK Fusion-Positive Tumors:
     
  • Adults: 600 mg once daily

     

  • Pediatric Patients (BSA-based):

     

    • BSA ≥1.51 m²: 600 mg

       

    • BSA 1.11–1.50 m²: 500 mg

       

    • BSA 0.91–1.10 m²: 400 mg

  • Continue until disease progression or unacceptable toxicity
  • Congestive Heart Failure: Monitor LVEF before and during treatment. Manage symptoms promptly.

  • CNS Effects: May include dizziness, cognitive changes, mood disturbance. Avoid hazardous activities if affected.

  • Fracture Risk: Monitor for signs of bone pain or mobility issues.

  • Liver Toxicity: Monitor AST/ALT levels biweekly during the first month, then monthly.

  • Hyperuricemia: Monitor serum uric acid. Initiate urate-lowering treatment if needed.

  • Pregnancy & Breastfeeding: May cause fetal harm. Use contraception during treatment and for 7 days after the last dose. Avoid breastfeeding during this period.
