Talzenna is a PARP inhibitor (poly [ADP-ribose] polymerase) indicated for the treatment of adult patients with germline BRCA-mutated (gBRCAm), HER2-negative, locally advanced or metastatic breast cancer.
Recommended Dosage:
- 1 mg orally once daily, with or without food
- If dose reduction is required, 0.25 mg capsules are available
- Continue until disease progression or unacceptable toxicity occurs
Administration Notes:
- Swallow capsules whole. Do not open, crush, or dissolve
- If a dose is missed or vomiting occurs, do not take an extra dose. Resume with the next scheduled dose
MDS/AML Risk: Myelodysplastic syndrome or acute myeloid leukemia may occur. Monitor CBC monthly. Discontinue if MDS/AML is confirmed
Myelosuppression: Monitor for leukopenia, anemia, neutropenia, or thrombocytopenia. Adjust dosage or withhold based on severity
Reproductive Toxicity: Talazoparib can cause fetal harm. Use effective contraception during and for 7 months after treatment
Breastfeeding: Avoid during treatment and for at least 40 days after the final dose
Pregnancy Testing: Required before starting treatment for females of reproductive potential
