Truseltiq (Infigratinib) is a kinase inhibitor indicated for adult patients with previously treated, unresectable locally advanced or metastatic cholangiocarcinoma with a fibroblast growth factor receptor 2 (FGFR2) fusion or rearrangement.

Recommended Dosage:

  • Standard Dose: 125 mg orally once daily (one 100 mg capsule + one 25 mg capsule) for 21 days, followed by 7 days off, in 28-day cycle

  • Administration: Take on an empty stomach (at least 1 hour before or 2 hours after food), at the same time daily. Swallow whole with water.
  • Missed Dose: If a dose is missed by ≥4 hours or vomiting occurs, skip the dose and resume the next day


Dosage Adjustments:

  • Mild/Moderate Renal Impairment: 100 mg once daily (21 days on/7 days off)

  • Mild Hepatic Impairment: 100 mg once daily

  • Moderate Hepatic Impairment: 75 mg once daily
  • Retinal Pigment Epithelial Detachment (RPED): May cause blurred vision. Perform OCT and eye exams at baseline, 1 month, 3 months, and every 3 months thereafter. Urgently refer if visual symptoms appear.

  • Hyperphosphatemia: May lead to mineralization or calcification of tissues. Regular phosphate level monitoring required. Initiate phosphate-lowering therapy as needed.

  • Pregnancy Risk: May cause fetal harm. Advise effective contraception during treatment and for 30 days after final dose (both males and females of reproductive potential).
