Zelboraf (vemurafenib) is an oral targeted therapy used in the treatment of BRAF V600 mutation-positive conditions. It is primarily prescribed for adults with:

  • Advanced or unresectable melanoma

  • Erdheim-Chester Disease (ECD) with BRAF V600 mutation


Vemurafenib works by inhibiting certain abnormal forms of the BRAF protein, which drive cancer cell growth.

Recommended Dosage:
The standard dosage is 960 mg (four 240 mg tablets) taken orally every 12 hours, with or without food. Treatment should continue until disease progression or the emergence of intolerable side effects.

  • Do not crush, chew, or split the tablets.

  • If a dose is missed, it may be taken up to 4 hours before the next scheduled dose.

  • If vomiting occurs, skip the dose and continue with the next scheduled one.
  • Secondary Malignancies: Vemurafenib increases the risk of developing new skin cancers, such as cutaneous squamous cell carcinoma. Monitor patients regularly for dermatologic changes.

  • Hypersensitivity: Severe allergic reactions may occur. Discontinue treatment if symptoms such as rash, fever, or swelling occur.

  • Serious Skin Reactions: Stevens-Johnson syndrome and other severe skin reactions have been reported. Discontinue therapy if severe rashes or blistering develop.

  • QT Prolongation: Vemurafenib may affect heart rhythm. Monitor ECG periodically and assess for signs like dizziness or fainting.

  • Ocular Toxicity: Eye inflammation and retinal disorders have been observed. Report any visual disturbances immediately.

  • Pregnancy Risk: This medication can cause fetal harm. Effective contraception must be used during treatment and for 2 weeks after the last dose.

  • Lactation: Breastfeeding is not advised during and for 2 weeks after the last dose of treatment, as effects on the infant are unknown.
